AN IMMUNOHIOSTOCHEMICAL STUDY ON THE CHANGE OF LAMININ IN BASEMENT MEMBRANE BY INVASION OF TUMOR CELL IN PRECANCEROUS LESION AND MALIGNANT EPITHELIAL TUMOR

Abstract

Invasion and metastasis are biologic hallmarks of malignant tumors. An important
attributes of metastatic cells is the ability to attach to, degrade, and penetrate the
extracellular matrix.
The purpose of this study was to evaluate the change of basement membrane by
invasion of malignant tumor cells by immunohistochemistry employing anti-laminin
primary antibody to the basement membrane.
12 cases of leukoplakia, 8 cases of carcinoma in situ, 20 cases of squamous cell
carcinoma, 7 cases of adenoid cystic carcinoma were reexamined and selected from the
files of the Department of Oral Pathology, College of Dentistry, Seoul National
University. Normal mucosa was obtained to serve as controls. Hematoxylin and Eosin
and PAS stained slides were prepared from formaline -fixed specimens. The Vectastain
Avidin-Biotin Complex(ABC) reagents were used for anti-laminin immunohistochemical
staining.
Following results were obtained.
1. The laminin staining in the basement membrane of normal oral mucosa and
leukoplakia showed continuous and regular linear patter.
2. The laminin staining in the basement membrane of carcinoma in situ showed almost
continuous and regular linear pattern, But thin, interrupted diffuse pattern in the
inflammatory cell infiltration area.
3. The laminin staining in the basement membrane of ell or moderately differentiated
squamous cell carcinoma showed interrupted, thin irregular pattern.
4. The laminin staining in the basement membrane of poorly differentiated squamous cell
carcinoma showed diffuse pattern or no response.
5. The laminin staining in the basement membrane in the tubular and cribriform pattern
of adenoid cystic carcinoma showed regular linear pattern, but no response around
neural sheath.
6. It was suggested that a specific antigenic defect of the basement membrane was
found in poorly differentiated squamous cell carcinoma and around neural sheath in
adenoid cystic carcinoma.
